La rogletimida , también conocida como piridoglutetimida , es un medicamento que nunca se comercializó. [ 1 ] Su estructura química es similar a la del fármaco sedante / hipnótico glutetimida , pero su actividad farmacológica es la de un inhibidor selectivo de la aromatasa, similar a la del fármaco relacionado aminoglutetimida , y carece de un efecto sedante-hipnótico significativo. [ 2 ] Esto la hace potencialmente útil en el tratamiento del cáncer de mama , con menos efectos secundarios que la aminoglutetimida, pero su menor potencia provocó su fracaso en los ensayos clínicos. Synthesis

Base catalyzed alkylation of ethyl 4-pyridylacetate [54401-85-3] (1) with iodoethane gives ethyl 2-(4-pyridyl)butyrate [76766-56-8] (2). Base catalyzed conjugate addition of the carbanion to acrylamide (3) gives (4). The last step is an intramolecular cyclization to rogletimide (5).
