La proteína de unión al determinante de la región codificante ( CRD-BP ) del ratón es una proteína de unión a ARN . [ 1 ] La CRD-BP pertenece a una familia de proteínas de unión a ARN que muestran una estrecha relación con la proteína de unión al código postal de β-actina del pollo ZBP1 [ 1 ] y las formas humanas de las proteínas IMP-1 , IMP-2 e IMP-3. [ 1 ] [ 2 ] [ 3 ] Debido a su estrecha relación, se cree que la CRD-BP y sus ortólogos comparten las mismas propiedades bioquímicas. Al unirse a sus transcritos, la CRD-BP participa en la traducción al estabilizar y localizar los transcritos en la célula. [ 3 ] Se ha observado una expresión normal de CRD-BP en el desarrollo temprano del embrión. [ 3 ] Por el contrario, la expresión de CRD-BP en el tejido adulto es extremadamente baja o completamente ausente. [ 4 ]

Estructura

CRD-BP es una proteína de 577 aminoácidos que contiene 4 dominios KH , 2 RRM y una caja RGG (Figura 2). [ 1 ] CRD-BP y la IMP-1 humana no son idénticas, pero muestran un alto grado de semejanza. Están muy relacionadas con IMP-2, lo cual se ha determinado mediante análisis filogenético (Figura 1). [ 3 ] Como resultado de esta estrecha filogenia , la estructura de los dominios funcionales conservados es compartida entre CRD-BP, ZBP1 y las IMP humanas (Figura 2). [ 3 ] [ 5 ] Los dominios KH conservados son compartidos en todos los ortólogos y pueden formar dímeros que orientan las regiones de unión al ARN de manera polarmente opuesta (Figura 3A). [ 5 ] Esta orientación crea espacio para que cada motivo GXXG (Figura 3B) de cada dominio KH se una a su transcrito de ARN (Figura 3A). [ 5 ]

Transcripciones de ARN de CRD-BP

CRD-BP and its orthologs have been shown to have the ability to bind CD44, beta-actin, c-myc, IGF2, H19 and tau RNA transcripts.[6] CRD-BP has been shown to play a crucial role in cancer growth and invasion of tissues. CD44 proteins belong to a family of cell surface adhesion molecules that are involved in both cell-cell and cell-matrix communications.[7] CRD-BP binds to and protects CD44 RNA which shows increased expression in cancers.[6] CRD-BP plays a role in the localization of β-actin mRNA by binding to the localization element in the 3'UTR region.[8] Aberrant expression of the oncogenic c-myc gene has been shown in the formation of tumours.[1] c-myc mRNA contains a coding region instability determinant (CRD) which CRD-BP has been shown to bind, therefore, protecting c-myc mRNA from endonucleolytic attack.[1] CRD-BP also binds IGF2 and H19 mRNA.[8] The H19 gene is located downstream of the IGF2 on chromosome 11 and chromosome 7 on humans and mice, respectively.[8] The binding of CRD-BP to these transcripts has shown to alter the expression of the genes. The microtubule-associated protein (MAP) is coded for by tau mRNA and is mainly found in the axon of neurons.[8] The 3’UTR of tau mRNA contains a cis-regulatory element that controls the axonal localization of tau mRNA.[8] CRD-BP binds to the axonal localization signal (ALS) of tau and plays a role in the localization of the transcript.[8]

CRD-BP, therefore, has been shown to be upregulated in many diseases, including cancers.
